Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated with BD Horizon BUV563 under optimum conditions, and unconjugated antibody and free BD Horizon BUV563 were removed.

Recommended Assay Procedures

For optimal and reproducible results, BD Horizon Brilliant Stain Buffer should be used anytime two or more BD Horizon Brilliant dyes are used in the same experiment. Fluorescent dye interactions may cause staining artifacts which may affect data interpretation. The BD Horizon Brilliant Stain Buffer was designed to minimize these interactions. R35-38

The R35-38 hybridoma produces monoclonal rat IgG2b, κ . The target molecule specificity of this clone is unknown. R35-38 has little or no reactivity with mammalian cells and has been found useful as an immunoglobulin isotype control.

The antibody was conjugated to BD Horizon BUV563 which is part of the BD Horizon Brilliant™ Ultraviolet family of dyes. This dye is a tandem fluorochrome of BD Horizon BUV395 which has an Ex Max of 348 nm and an acceptor dye. The tandem has an Em Max at 563 nm. BD Horizon BUV563 can be excited by the 355 nm ultraviolet laser. On instruments with a 561 nm Yellow-Green laser, the recommended bandpass filter is 585/15 nm with a 535 nm long pass to minimize laser light leakage. When BD Horizon BUV563 is used with an instrument that does not have a 561 nm laser, a 560/40 nm filter with a 535 nm long pass may be more optimal. Due to the excitation and emission characteristics of the acceptor dye, there may be spillover into the PE and PE-CF594 detectors. However, the spillover can be corrected through compensation as with any other dye combination.

BD Horizon™ BUV563 is part of the BD Horizon Brilliant™ Ultraviolet family of dyes. This tandem fluorochrome is comprised of a BD Horizon BUV395 donor with an excitation maximum (Ex Max) of 350 nm and an acceptor dye with an emission maximum (Em Max) at 564 nm. BD Horizon BUV563, driven by BD innovation, is designed to be excited by the ultraviolet laser (355 nm) and detected using an optical filter centered near 560 nm (eg, a 560/40 or a 585/15 nm bandpass filter). The acceptor dye can be excited by the Blue (488 nm) laser resulting in cross-laser excitation and fluorescence spillover. Please ensure that your instrument’s configuration (lasers and optical filters) are appropriate for this dye.
