Smart Panel Design
SMART Panel Design Lowers Sequencing Cost While Preserving the Sensitivity and Resolution of CITE-Seq Data
CITE-seq or high-plex protein-profiling experiments with antibody-oligos traditionally incur high sequencing costs because highly abundant protein targets such as CD45 and CD44 consume most of the sequencing reads requiring deeper sequencing to detect less abundant secondary and tertiary protein targets. SMART panel design addresses this challenge by utilizing carefully selected, pre-titrated concentrations of antibody-oligos against abundant primary markers. This helps to distribute sequencing reads to the more critical secondary and tertiary targets without compromising the sensitivity or ability to detect the abundant cell surface primary markers.
How does this work? Select antibody-oligos in each BD® OMICS-One Protein Panel that target select, highly expressed primary markers are titrated and lyophilized alongside “cold antibodies” (antibodies lacking oligonucleotides) of the same clone. This approach generates a gradual and predictable decrease in reads allocated to the primary marker targeted thereby moderating the signal of the targeted marker without affecting the ability to resolve cell populations. The outcome is better resolution of low-expressed proteins at the same sequencing depth without incurring additional sequencing costs.
