BD Horizon™ RB545 Reagents offer minimal cross-laser excitation off the yellow-green laser
Applications
RB545 reagents can resolve high-expression surface and intracellular markers and are available in a wide array of specificities. RB545 reagents can be used with PE and FITC on spectral flow cytometers to meet your specific panel design needs.
RB545 reagents can help improve resolution in high-parameter spectral flow cytometry panels.
Human PBMCs were stained with two sets of reagents, moderately similar CD4 RB545 and CD127 FITC, and highly similar CD4 FITC and CD127 BB515. Co-expressed markers were selected to demonstrate impact of spectral unmixing with different levels of similarity. Resolution was impacted when reagents were highly similar, shown in bottom set of CD4 FITC and CD127 BB515. Samples were acquired on the BD FACSymphony™ A5 SE Cell Analyzer and spectrally unmixed with FlowJo™ Software.
RB545 reagents show stable performance with lot-to-lot consistency across made-to-stock reagents and BD OptiBuild™ On-Demand Reagents. They also offer proven photostability when exposed to typical lab lighting or dimmed core lab lighting.

BD Horizon RealYellow™ 586 (RY586) Reagents are now available as the first in the rapidly growing family of innovative, laser-specific fluorochromes. They are excited primarily by the 561-nm yellow-green laser and offer:

RY586 can be used with PE on spectral flow cytometers. Since RY586 provides comparable brightness to PE, it is ideal for detecting low-expression markers. Using RY586 and PE in the same spectral flow cytometry panel enables high-dimensional analysis of more low-expression surface and intracellular markers.

Performance stability of BD Horizon RealYellow™ Reagents RY586 offers stable reagent performance with lot-to-lot consistency across made-to-stock reagents and BD OptiBuild™ On-Demand Reagents as well as proven photostability when exposed to typical lab lighting or dimmed core lab lighting.
RY586 made-to-stock reagents and BD OptiBuild™ On-Demand Reagents show consistent performance between each other and across multiple lots.
Human whole blood was stained with human CD4 (SK3) RY586, followed by lysis with BD FACS™ Lysing Solution. Mouse splenocytes were stained with mouse CD8a 53-6.7. All specificities were run on a BD FACSymphony™ A5 SE Cell Analyzer.

APPLICATIONS
The BD FACSymphony™ A5 Cell Analyzer enables high-parameter analysis for a wide array of applications, including phenotyping of human blood, analysis of murine leucocyte subsets and assessment of surface markers of human ILC1 cells.
Human PBMCs were stained with a 25-color immunophenotyping panel and analyzed on the BD FACSymphony™ A5 Cell Analyzer. Five different healthy donors each were run on the analyzer with identical instrument configurations at three different sites. Donor 1 data are shown here.
Comparable segregation of murine cell populations defined by a 28-color reagent panel on the BD FACSymphony™ A5 Cell Analyzer. Two-dimensional t-SNE map generated after merging cells from bone marrow, blood, spleen and lymph nodes. Cell populations were manually gated and overlaid onto the t-SNE plot. Doughnut charts display the frequency of the indicated cell populations in each individual tissue.
Fresh PBMCs were stained with a 21-color panel and analyzed on the BD FACSymphony™ A5 Cell Analyzer. Lineage CD3-CD56-CD127+CD117-CD294- ILC1 cells were gated into CD4-CD8- (blue), CD4+CD8- (red) and CD4-CD8+ (green) subsets and further analyzed for protein expression patterns.

Automated sample preparation and antibody cocktail preparation to improve efficiency
Flow cytometry sample preparation can be automated using the BD FACSLyric™ Flow Cytometer integrated with the BD FACSDuet™ Premium Sample Preparation System. The sample preparation system is designed to complement and physically integrate with the flow cytometer to provide a complete end-to-end walkaway solution. This system helps streamline workflow and drive productivity while allowing for on-board automated antibody cocktail preparation which eliminates risk of errors due to manual pipetting. It supports different types of blood collection tubes and a wide variety of reagent vials delivering flexibility. The BD FACSLyric™ Flow Cytometer enables efficiency and productivity with automated laser alignment and compensation stability over 60 days and allows flexibility for acquisition using a universal loader with 21 different loading options including multiwell plates. Contract manufacturing of ready-to-use multicolor reagents panels in liquid, dried, or lyophilized formats are customized to your specifications and requirements. Dried and lyophilized reagents are manufactured using proprietary BD Horizon™ Dri Chroma Reagent technology and BD Horizon™ Lyo Reagent technology, respectively and have extended stability compared to liquid reagents. All-in-one tube format reduces the need for manual pipetting and cocktailing which improves consistency, reduces operator errors and day-to-day variability and saves time.
